Sifting does make a difference, and I always use a fine mesh strainer. When sifting flour, I also add my other dry ingredients to the top of the flour pile, and it combines the ingredients very well, as well as catches any clumps that have stuck together, such as in baking soda, cocoa, salt, etc.
