Lowry Protein Assay

The Lowry Protein Assay is the first of its kind of scientific method to determine the total protein content of a given sample. The method applies the molecular reactions of copper ions with the protein peptide bonds. Resulting to the oxidation of aromatic protein residues under alkaline conditions. One disadvantage of using this method is the delayed incubation time of measuring the total protein concentration. The method was first described in an original publication entry entitled "Protein measurement with the Folin phenol reagent" by Lowry, OH and et. al. in the Journal of Biological Chemistry last November 1951. Since then the method has been modified and improved to suit the protein-to-protein variation during total protein concentration measurement.

Modified Lowry Protein Assay (Hancock Lab); The advantage of this assay over the Lowry assay from which it is derived, is the presence of SDS in reagent A. This allows one to perform protein assays in nearly any detergent (eg., when detergents are used in the purification of outer membrane proteins).
