Dear All, I'm trying to infect PBMCs with some mycobacterial strains at a specified multiplicity of infection (MOI). How can I determine the MOI? Please advise. I have no experience with kind of work. Your advise and comments will be highly valued.

The multiplicity of infection (abbreviated MOI) is the average number of phage per bacterium. The MOI is determined by simply dividing the number of phage added (ml added x PFU/ml) by the number of bacteria added (ml added x cells/ml).

Infect cells at a multiplicity of infection (MOI) 5-10 (i.e., at 5-10 pfu/cell). For example, if the T175 flask contains ~ 18 x 10 6 cells, add 1.8 x 10 8 pfu adenovirus to get an MOI of 10 pfu/cell. Dilute the virus with media if necessary.
