Cystinuria is a genetic defect in the kidney tubules. Normal kidneys filter the amino acid cystine and do not allow it to go into the urine. In dogs with cystinuria, this filtering action fails, and cystine passes into the urine, where it can form crystals and/or stones (uroliths).

Although cystinuria affects many dog and cat breeds, the most severe form seems to affect Newfoundland dogs. Cystinuria is inherited as an autosomal recessive trait in the Newfoundland dog. In other words, affected Newfoundland dogs have two mutant genes, one inherited from each parent.

Cystinuria in Newfoundland dogs is a metabolic disease associated with a nonsense mutation in the exon 2 of the Slc3a1 gene. Similar to type I human cystinuria, heterozygote carriers are not affected by the disease and do not reveal differences in urinary concentration of dibasic amino acids when compared with normal dogs.
