DNA replication is initiated at a region on a chromosome called an origin of replication. An enzyme called T-antigen (helicase) binds to the origin and unwinds the DNA in both directions from the origin.

DNA replication is semi-conservative, one strand serves as the template for the second strand. Furthermore, DNA replication only occurs at a specific step in the cell cycle. The following table describes the cell cycle for a hypothetical cell with a 24 hr cycle.

This is the stage when DNA replication occurs. The G2 stage stands for "GAP 2". The M stage stands for "mitosis", and is when nuclear (chromosomes separate) and cytoplasmic (cytokinesis) division occur. Mitosis is further divided into 4 phases, which you will read about on the next page.

What does a chromosome look like following DNA rep ... What does a chromosome look like following DNA replication? What happens during the M stage of the cell cycle? What effect would be seen in the daughter cells if cytokinesis did not occur after mitosis? If sister chromatids were not separated during mitosis?

DNA replication occurs during interphase. The DNA must be copied in preparation for cell division (mitosis and meiosis). Specialized cells like muscle and nerve cells do NOT divide and therefore do NOT replicate their DNA.

The processing of concatemers to produce mature viral DNA and virus assembly also occurs in the cytoplasm. This mode of replication is strikingly different from any other known DNA virus.
