The coarse adjustment knob is the first device for focusing. With the scanner objective (4X) in place, this knob is used to determine a sharp focus. All other objectives should be in "near focus" when rotated into position and will only need the fine adjustment for a sharp image.

Each part of a microscope is identified and it's operation explained ... Once the image is sharp with the low power lens, you should be able to simply click in the next power lens and do minor adjustments with the focus knob. If your microscope has a fine focus adjustment, turning it a bit should be all that's necessary.
