Each nucleosome is composed of DNA wrapped around an octamer of the four core histones H2A, H2B, H3 and H4. This tight packaging, which is further helped by linker histones and other less-abundant proteins, produces condensed chromosomes that allow faithful segregation of the genetic material in dividing cells.

Since opposite charges attract, the DNA is pulled in toward the histones. A nucleosome is a segment of the DNA wrapped around a core of histones.

Current models of the yeast kinetochore propose that the yeast centromere DNA is wrapped around a Cse4p variant nucleosome. Here we present evidence that the histone-fold domain of Cse4p interacts specifically with CDE I and CDE II centromere DNA.

Eukaryotic chromosomes are packaged in condensed chromatin structures whose primary unit, the nucleosome, is composed of about 147 DNA base pairs wrapped around a histone octamer (1,2). Nucleosomes, as well as the enzymes that remodel and modify them, are key regulators of genome activity (3).
