Where gap junctions are abundant, membranes stain darker and histologists named these areas intercalated disks. Cardiac myofibers also contain a single, centrally located nucleus in contrast to the multiple peripheral nuclei of skeletal muscles.

Intercalated disks appear as slightly darker lines perpendicular to the length of the cardiac muscle fibres. Depending upon the preparation and the staining, intercalated disks can be obvious or barely identifiable.

CARDIAC MUSCLE CELLS WITH INTERCALATED DISKS ... The intercalated (arrows) discs are specialized cell junctions between CMF’s. They are located At Z lines and carry out important functions for the cell. ... The disc contains gap junctions which permit the rapid conduction of chemical and electrical activities between CMF’s.

Electron microscopy revealed the presence of nascent intercalated disks connecting the engrafted fetal cardiomyocytes and the host myocardium. These results suggest that intracardiac grafting might provide a useful approach for myocardial repair, provided that the grafted cells can contribute to myocardial function.

β-Catenin accumulates in intercalated disks of hypertrophic cardiomyopathic hearts ... Results: Intercalated disks are disorganized in both hamster and human cardiomyopathic hearts; β-catenin is increased and accumulated in intercalated disks depriving cardiomyocyte nuclei of fundamental signals.
