Methyl Red (MR) and Voges-Proskauer (VP) broth is used as a part of the IMViC tests as the medium in which both the Methyl Red and Voges-Prosakuer tests can be performed. It is a simple broth that contains peptone, buffers, and dextrose or glucose.

The Methyl-Red test tests for the ability to perform mixed-acid fermentation. MR-VP broth contains glucose, peptone, and a phosphate buffer. Organisms that perform mixed-acid fermentation produce enough acid to overcome the buffering capacity of the broth, so a decrease in pH results.

Methyl red shows its acid colour, red, if the pH is 4.2 or lower, and is completely in its alkaline (yellow) form when the pH reaches 6.3. It should show its intermediate orange colour at around pH 5.2. Have you tried checking the pH at the start, or are you using a reagent that is too alkaline to begin with?
