Inoculating needle is used like a pen. Hold it like you hold a pen. Inoculating loop and a needle is mainly used to pick a single colony(pure) so u need to be gentle on the agar. practice using an inoculating needle on a paper with pen.

Explain why a straight inoculating needle is used to inoculate an agar deep tube?...that one i have no ide ... You're correct about the first two. Though the contamination doesn't just come from the air. The stopper may have some contamination as well. The straight needle allows for uniform innoculation.

(Actually, an inoculating needle should be used rather than an inoculating loop to better visualize motility in the medium; however, the needles are not available for this experiment).  The stabs are incubated at 35oC in ambient air for 48-72 hours with the caps slightly loosened.
