Light microscopy does not reveal any substructure in the elastic fibres. Electron microscopy shows that elastic fibres consist of individual microfibrils, which are embedded in an amorphous matrix. The matrix accounts for about 90% of the fibre and is composed of the protein elastin.

These cells secrete the components which make up the fibres and ground substance that form the non-cellular component of the loose connective tissue. In the slide preparation above, both collagen fibres (1 - bundle of fibres circled) and elastic fibres (2 - each a single fibre) can be seen.
